Intonation refers to whether a note is in tune. On a fretless bass, attention must be given to the placement of your fingers to ensure that notes are not sharp or flat.

The main point about intonation is that you will need to hold the notes a little closer towards the bridge end of the neck. This is because on a fretted bass your fingers were behind the fret but now they need to be right on where an imaginary fret would have been.

Opinion is divided as to whether it is cheating to use a fretless bass with fret lines which mark where the frets would have been. I don't see how it matters as music is about the sounds you make and not how difficult something is. It's also ironic when you consider that Jaco Pastorius, the most highly regarded bass player of all time, had fret markings as he actually removed the frets himself from a Fender Jazz bass which left lines on the neck where the frets had been.
